In cucumber, the genetic basis of traits under domestication and/or diversifying selection is not well understood. Here, we reported QTL mapping for flowering time and fruit size-related traits with segregating populations derived from a cultivated × wild cross. Phenotypic data of flowering time (FT), fruit size (FS), fruit number (FN) and fruit weight per plant (FW) were collected in multiple environments. QTL analysis identified 19 QTL for these traits. We found that the major-effect QTL FT1.1 played an important role in regulating flowering time in cultivated cucumber, whereas the minor-effect QTL FT6.3 contributed to photoperiod sensitive flowering time during domestication. Two novel consensus FS QTL, FS1.4 and FS2.3, seem to be the targets of selection during breeding for the US processing cucumber. All other FS QTL were co-localized with previously detected QTL using populations derived from cultivated cucumbers, suggesting that they were under selection during both initial domestication and subsequent improvement. Results from this study also suggested that the wild cucumber is a useful resource for capturing positive transgressive segregation and novel alleles that could be explored in cucumber breeding. 相似文献

Six new monosulfated triterpene tetra-, penta- and hexaosides, namely, the kurilosides A1 (1), A2 (2), C1 (3), D (4), E (5) and F (6), as well as the known earlier kuriloside A (7), having unusual non-holostane aglycones without lactone, have been isolated from the sea cucumber Thyonidium (= Duasmodactyla) kurilensis (Levin) (Cucumariidae, Dendrochirotida), collected in the Sea of Okhotsk near Onekotan Island from a depth of 100 m. Structures of the glycosides were established by 2D NMR spectroscopy and HR-ESI mass spectrometry. Kurilosides of the groups A and E contain carbohydrate moieties with a rare architecture (a pentasaccharide branched by C(4) Xyl1), differing from each other in the second monosaccharide residue (quinovose or glucose, correspondingly); kurilosides of the group C are characterized by a unique tetrasaccharide branched by a C(4) Xyl1 sugar chain; and kurilosides of the groups D and F are hexaosides differing from each other in the presence of an O-methyl group in the fourth (terminal) sugar unit. All these glycosides contain a sulfate group at C-6 of the glucose residue attached to C-4 Xyl1 and the non-holostane aglycones have a 9(11) double bond and lack γ-lactone. The cytotoxic activities of compounds 1–7 against mouse neuroblastoma Neuro 2a, normal epithelial JB-6 cells and erythrocytes were studied. The system nitrogen (N), phosphorous (P) budget, N/P utilization rate and the physiological response mechanism of the abalone Haliotis discus hannai (body weight: 12.87 ± 0.82 g) and the sea cucumber Apostichopus japonicus (body weight: 10.85 ± 1.16 g) to different co‐culture environment conditions were examined. Animals were kept in a multilayer, cubic recirculating aquaculture system at different polyculture densities (abalones at 400 ind/m2in monoculture [Group C] and abalones at 400 ind/m2with sea cucumbers at 10 ind/m2 [AS1] or 20 ind/m2 [AS2]). Each treatment was replicated four times, and the experimental cycle was 90 days. No significant difference in survival rate of abalones was detected when the stocking density of sea cucumbers increased from 10 to 20 ind/m2, but the concentrations of total ammonia nitrogen and nitrite nitrogen in the water were significantly higher in AS2 than in AS1. Survival rate, specific growth rate (SGR) of body weight of sea cucumbers and SGR of body weight of abalones were significantly lower in AS2 than in AS1. No significant difference in protease (PES), lipase, amylase and cellulase activities of abalones was identified between Group C and AS1, but the PES and amylase activities of abalones and sea cucumbers in AS1 were significantly higher than those in AS2. In AS1, the N/P output from harvesting of abalones and sea cucumbers and the N/P utilization rates were significantly higher than those in AS2. Although the N/P output from faeces was significantly lower in AS2 than in Group C and AS1, the N/P output from the water layer was significantly higher than that in AS1. The expression levels of Cu/Zn superoxide dismutase (Cu/Zn‐SOD) and heat shock protein 70 (HSP70) of both abalones and sea cucumbers in AS2 were significantly higher than those in AS1. No significant difference in expression of catalase (CAT) and HSP90of abalones was identified among these groups, but the expression levels of CAT and HSP90of sea cucumbers in AS2 were significantly higher than those in AS1. These results indicate that stocking sea cucumbers at 10 ind/m2 in the polyculture system will relieve the organic load on the system and improve the N/P utilization rate. 为探明基质栽培黄瓜适宜的灌水下限,实现农艺节水,以黄瓜品种博特209为试材,采用基质盆栽,共设置4个灌水下限处理,即田间持水量的50%、60%、70%、80%,用A、B、C、D表示,灌水上限统一设定为田间持水量的90%,采用TDR350水分速测仪控制基质的水分含量,研究不同灌水下限对基质栽培黄瓜叶片水分状况、光合日变化及荧光参数的影响。结果表明:随着灌水下限的提高,叶片的相对含水量、自由水含量及自由水/束缚水的比值呈逐渐上升的趋势,水分饱和亏、叶片细胞汁液浓度及束缚水含量则呈下降趋势,处理C、D的相对含水量、自由水含量均显著(P<0.05)高于处理A、B,处理A细胞汁液浓度显著高于处理C、D,处理A、B束缚水含量显著(P<0.05)高于处理C、D。叶绿素a、叶绿素b和叶绿素a+b的含量均随灌水下限的升高呈现为先上升后下降的趋势,而类胡萝卜素含量则表现为先下降后上升的趋势,处理C叶绿素a、叶绿素a+b含量显著(P<0.05)高于处理A、B;处理C、D黄瓜叶片的胞间二氧化碳浓度(Ci)的变化趋势基本呈“V”型,而处理A、B的Ci的变化趋势基本呈“W”型;处理A、B、C、D的蒸腾速率(Tr)日变化均呈先上升后下降的趋势,处理B、C、D的Tr在14:00后呈现逐渐下降的趋势,而处理A在12:00后就呈快速下降的趋势;处理B、C、D的气孔导度(Gs)日变化大致呈“M”型的双峰曲线,处理A则呈单峰曲线;处理A、B、C、D叶片净光合速率(Pn)与Gs日变化趋势基本一致,在整个日变化过程中,处理C、D的Pn始终高于处理A、B。Fv/Fm、ΦPSⅡ、qP随灌水下限的提高呈逐渐上升的趋势,而NPQ呈逐渐下降的趋势。研究结果表明,灌水下限为田间持水量的70%~80%,基质栽培黄瓜叶片的水分状况良好、光合色素含量高、净光合速率大,有利于黄瓜的光合作用,促进黄瓜的生长发育。 相似文献

High water consumption and inefficient irrigation management in the agriculture sector of the middle and lower reaches of the Amu Darya River Basin(ADRB)have significantly influenced the gradual shrinking of the Aral Sea and its ecosystem.In this study,we investigated the crop water consumption in the growing seasons and the irrigation water requirement for different crop types in the lower ADRB during 2004–2017.We applied the FAO Penman–Monteith method to estimate reference evapotranspiration(ET0)based on daily climatic data collected from four meteorological stations.Crop evapotranspiration(ETc)of specific crop types was calculated by the crop coefficient.Then,we analyzed the net irrigation requirement(NIR)based on the effective precipitation with crop water requirements.The results indicated that the lowest monthly ET0 values in the lower ADRB were found in December(18.2 mm)and January(16.0 mm),and the highest monthly ET0 values were found in June and July,with similar values of 211.6 mm.The annual ETc reached to 887.2,1002.1,and 492.0 mm for cotton,rice,and wheat,respectively.The average regional NIR ranged from 514.9 to 715.0 mm in the 10 Irrigation System Management Organizations(UISs)in the study area,while the total required irrigation volume for the whole region ranged from 4.2×109 to 11.6×109 m3 during 2004–2017.The percentages of NIR in SIW(surface irrigation water)ranged from 46.4%to 65.2%during the study period,with the exceptions of the drought years of 2008 and 2011,in which there was a significantly less runoff in the Amu Darya River.This study provides an overview for local water authorities to achieve optimal regional water allocation in the study area. 相似文献

BACKGROUND
Ecballium elaterium (common name: squirting cucumber) is an emerging weed problem in hedgerow or superintensive olive groves under no tillage. It colonizes the inter-row area infesting the natural or sown cover crops, and is considered a hard-to-control weed. Research in other woody crops has shown E. elaterium has a patchy distribution, which makes this weed susceptible to design a site-specific control strategy only addressed to E. elaterium patches. Therefore, the aim of this work was to develop a methodology based on the analysis of imagery acquired with an uncrewed aerial vehicle (UAV) to detect and map E. elaterium infestations in hedgerow olive orchards.RESULTS
The study was conducted in two superintensive olive orchards, and the images were taken using a UAV equipped with an RGB sensor. Flights were conducted on two dates: in May, when there were various weeds infesting the orchard, and in September, when E. elaterium was the only infesting weed. UAV-orthomosaics in the first scenario were classified using random forest models, and the orthomosaics from September with E. elaterium as the only weed, were analyzed using an unsupervised algorithm. In both cases, the overall accuracies were over 0.85, and the producer's accuracies for E. elaterium ranged between 0.74 and 1.00.CONCLUSION
These results allow the design of a site-specific and efficient herbicide control protocol which would represent a step forward in sustainable weed management. The development of these algorithms in free and open-source software fosters their application in small and medium farms. © 2022 The Authors. 采用平板计数法和聚合酶链式反应( PCR)-变性梯度凝胶电泳( DGGE)技术分析免疫增强剂党参对仿刺参( Apostichopus japonicas)肠道菌群结构的影响。将初始体重为(18.00±2.00) g的仿刺参随机分为2组(对照组、试验组),每组6个重复,每个重复12只。对照组投喂海泥、鼠尾藻粉按照1∶1的质量比配制的饵料,试验组饵料中以鼠尾藻粉质量的2%添加党参,连续投喂28 d。结果表明:应用免疫增强剂党参不仅能够显著提高仿刺参的特定生长率( P<0.05),降低其饵料系数(P<0.05),而且能够显著增加仿刺参肠道内容物中异养菌的数量(P<0.05);序列统计分析显示投喂党参后仿刺参肠道细菌优质序列比例显著增加( P<0.05),试验组达97.57%,对照组仅为80.22%;Beta多样性分析反映投喂党参后仿刺参肠道微生态环境发生了变化,其多样性系数范围在14.91%~15.47%、15.47%~16.21%、14.91%~16.21%;丰度分析显示投喂党参后仿刺参肠道内容物中变形菌门( Proteobacteria)和拟杆菌门( Bacteroidetes)丰度提高,疣微菌门( Verrucomicrobia)、放线菌门( Actinobacteria)和厚壁菌门( Firmiaites)丰度降低;聚类分析显示试验组与对照组肠道菌群结构相似性系数为0.97。由此可见,免疫增强剂党参可提高仿刺参的特定生长率,降低饵料系数,增加肠道异养菌数量和优势菌群丰度,优化肠道微生态环境。 相似文献

从患病鲈鱼体内分离到1株优势菌LY2014,对其进行形态观察、生理生化试验、16SrDNA基因序列分析,同时采用Kriby-Bauer纸片扩散法进行耐药性分析。结果该菌为发酵型、具有动力的革兰阴性杆菌,根据生化试验结果初步确定该菌为摩氏摩根菌。以该菌的基因组为模板PCR扩增获得的16SrDNA序列长度为1 407bp(GenBank登入号为KJ830813)。该序列与GenBank数据库中摩氏摩根菌的16SrDNA基因序列相似性高达99%,系统进化树显示其与摩氏摩根菌亲缘关系最近,从而确定菌株LY2014为摩氏摩根菌。该菌株对磷霉素、菌必治和多黏霉素B高度敏感,对痢特灵、红霉素和诺氟沙星等5种药物中度敏感,对氨苄西林和氟苯尼考等11种药物耐药。 相似文献
